为培育一种蛋黄胆固醇含量较低的蛋鸡新品系,以丝羽乌骨鸡为研究对象,测定675枚丝羽乌骨鸡蛋蛋黄胆固醇含量,通过DNA测序技术和创造酶切位点PCRRFLP方法检测了丝羽乌骨鸡群体中3羟基3甲基戊二酸单酰辅酶A还原酶基因的2个SNPs位点,分别分析这两个SNPs位点不同基因型与鸡蛋胆固醇含量的关联性。结果表明:(1)丝羽乌骨鸡蛋蛋黄胆固醇含量为13.41 mg/g,鸡蛋中胆固醇含量为456.50 mg/100g。(2)g.12217G>T 位点中GG基因型的鸡蛋胆固醇含量最低,且与其他两种基因型具有显著的差异。
Abstract
In order to foster a low yolk cholesterol new lines of laying hens,675 yolk cholesterol of Silky Egg were measured.DNA sequencing technology was adopted to create restriction site PCRRFLP and detect HMGCR SNPs in the silkies, and were the correlation between the two SNPs in different genotypes and egg cholesterol content analyzed. Based on association analysis of these HMGCR polymorphisms in the silkies, it's found that Silky Egg yellow cholesterol content is 13.41 mg/g, and the cholesterol content in eggs is 456.50 mg/100g.The g.12217G>T site showed associations with the cholesterol content in eggs, and the cholesterol content in eggs of genotype GG was the lowest (P<0.1).
