摘要

雌激素相关受体α(Estrogen-related receptors α,ESRRα)是一个多功能蛋白,主要参与全身能量代谢,并且在脂肪细胞分化过程中起着重要作用。研究采用直接测序与限制性内切酶片段长度多态性 (RFLP) 相结合的方法,对282头秦川牛ESRRα基因的第一内含子进行多态性检测,发现一个单核苷酸多态位点(SNP)(C147A)。通过分析得出:在这个秦川牛群体中存在2个等位基因,A和C,两个等位基因的频率分别为0.4025和0.5975,检测到3种基因型,AA、CA和CC,3种基因型的频率分别为0.1844,0.4363和0.3794。ESRRα基因与秦川牛肉质性状的关联分析结果表明:CC基因型对牛的背膘厚有较大影响 (P<0.05);而AA基因型和CA基因型个体的大理石花纹则显著高于CC型个体 (P<0.05)。

Abstract

Estrogen-related receptors α (ESRRα) is a multifunctional protein which is involved in differentiation of preadipocytes to adipocytes and body energy metabolism. It's an important candidate gene for meat quality selection through markerassisted selection (MAS). In this study, based on the PCR-RFLP technology,one single nucleotide polymorphism (SNP) (C147A in inxon 1) of the ESRRα gene was discovered. The analysis result showed that there were 2 alleles (A and C) and 3 genotypes(AA,CA and CC) in 282 Qinchuan Cattle.The frequences of alleles found was 0.4025 for A and 0.5975 for C. The frequences of genotypes found was 0.1844 for AA,0.4363 for CA and 0.3794 for CC.The association between genotype markers and meat traits were analyzed by general linear model at the level of DNA. The result was the CC genotype of Qinchuan CattleESRRα gene showed a highly significant negative effection Marbling (MB )traits (P< 0.05 ) ,and animals with the CC genotype have a significantly lower Backfat thickness (BFT) compared to the CA and AA genotypes (P<0.01).
