清瘟败毒颗粒中黄连和连翘的薄层鉴别

摘要

薄层层析方法用于清瘟败毒颗粒中黄连和连翘的鉴别。试验以苯-乙酸乙酯-甲醇-异丙醇-浓氨溶液(12∶6∶3∶3∶1)为展开剂,置氨蒸气饱和的展开缸内展开,于紫外灯下检视黄连和盐酸小檗碱的存在。以三氯甲烷-甲醇(5∶1)为展开剂,10%硫酸乙醇溶液为显色剂,105 ℃烘5 min,自然光下检测连翘的存在。结果表明,在365 nm紫外灯下,清瘟败毒颗粒与黄连对照药材、盐酸小檗碱在Rf值相同处有相同斑点;在自然光下,清瘟败毒颗粒与连翘对照药材在Rf值相同处有相同斑点。建立了清瘟败毒颗粒中黄连和连翘的薄层鉴别方法,专属性强,重现性好,适合于快速检验。

Abstract

TLC method was used for detection of Coptidis Rhizama and Forsythia in Qingwen Baidu Keli. The proportion of Benzene ethyl acetate methanolisopropanolstronger ammonia water (12∶6∶3∶3∶1) was used as the mobile phase, put into the cylinder with saturation of ammonia steam to outspread, and detect the existence of Coptidis Rhizama under UV lamp. The proportion of Chloroform-Methanol (5∶1) was used as the mobile phase, the 10% sulfuric acid ethanol solution was colour reagents, and heated to 105 ℃ for 5 minutes to detect the existence of Forsythia with natural light. The results show there was same spots at the same Rf value between Qingwen Baidu Keli, Rhizoma Picrorhizae and Berberine hydrochloride under 365 nm UV lamp, there was also same spots between Qingwen Baidu Keli and Fructus Forsythiae under natural light. This method is simple and special in operation, the results are stable with a good repeatability. These can be well applied to control the quality of Qingwen Baidu Keli.
